在ROC分析中,Ribeye对SNHL的判别性能优于Prestin(曲线下面积[AUC],0.76 vs. 0.65)。

PURPOSE: The aim of this study was to evaluate serum Prestin and Ribeye (CtBP2) levels in children with bilateral congenital profound sensorineural hearing loss (SNHL) and to investigate their potential as circulating biomarkers of cochlear pathology.
METHODS: A case-control study was conducted at a tertiary referral otorhinolaryngology center. A total of 153 children aged 1-6 years were included, comprising 75 children with bilateral congenital profound SNHL and 78 normal-hearing controls. Serum Prestin and Ribeye concentrations were measured in peripheral blood samples using enzyme-linked immunosorbent assay (ELISA). Diagnostic performance was assessed using receiver operating characteristic (ROC) analysis, and associations with hearing loss status were evaluated using multivariate analysis of covariance (MANCOVA) and logistic regression analysis.
RESULTS: Serum Prestin and Ribeye levels were significantly higher in children with congenital profound SNHL than in normal-hearing controls (median Prestin: 930.70 vs. 414.08 pg/mL, p = 0.001; median Ribeye: 15.49 vs. 6.26 ng/mL, p < 0.001). After simultaneous adjustment for age and sex using MANCOVA, the overall biomarker profile remained significantly associated with hearing status (Wilks' λ = 0.865, p < 0.001). In ROC analysis, Ribeye demonstrated greater discriminatory performance for SNHL than Prestin (area under the curve [AUC], 0.76 vs. 0.65). Logistic regression analysis showed that each 1-unit increase in serum Ribeye level was associated with an 18% increase in the odds of SNHL (odds ratio, 1.18).
CONCLUSION: Children with congenital profound SNHL exhibited higher serum levels of Prestin and Ribeye than normal-hearing controls. While both biomarkers showed potential, their performance may not yet be sufficient to support their use as standalone clinical tests. Further longitudinal studies with age-matched controls and direct biological validation are required to determine whether these circulating proteins could complement existing audiological assessments of congenital hearing loss.
